| The crash, involving a green Citroen Xantia and a white Fiat van, occurred on a straight stretch of the A915 Standing Stane road that runs between Kirkcaldy and Leven at around 7.15 am.
Emergency services and paramedics arrived at the scene within a few minutes of the first 999 calls.
However, neither of the casualties, who were both in their 30s, could be saved.
The route was closed between the Boreland and Check Bar roads as the casualties were removed and an investigation was launched by police.
The car had been travelling west towards Kirkcaldy and the van driving in the opposite direction.
The men who died were the sole occupants of their vehicles.
The A915 was closed for most of the morning causing delays throughout the area.
The identities of the two men will not be released until relatives have been informed, say police.
Inspector Mike Stevens said, “This investigation is at an early stage but we can state that an overtaking manoeuvre was not a contributory factor in this tragic accident.”
